The test tube: the perfect founding setup

The test tube is the universal setup for housing a mated queen. Sturdy, simple to make and cheap, it’s the essential first step of any colony start-up. Here is the proven method.

What you need

• 1 glass test tube 16×150 mm (standard size)
• Untreated cotton wool (unscented, unbleached)
• Filtered or still mineral water
• A syringe or dropper (optional but handy)
• Foil or black fabric for darkness

Step 1: fill the water reservoir

Pour in about 1/4 of the tube with water, i.e. 2 to 3 cm deep. The water should cover the closed end of the tube.

Step 2: the wet cotton plug

Prepare a well-packed cylinder of cotton wool that pushes 2-3 cm into the tube. Gently push it in with a pencil or stick until it touches the water. The cotton soaks up the water and creates a watertight barrier.

Step 3: the main chamber

The space above the wet cotton is the founding chamber. This is where the queen will live and found her colony. This space is typically 10-12 cm long.

Step 4: introducing the queen

Introduce the queen through the open end of the tube using soft tweezers or a brush. Blow gently to encourage her in. Once inside, she’ll naturally head toward the wet cotton.

Step 5: the exit plug

Plug the open end with a cylinder of dry cotton (not wet) that closes the tube well. This plug lets a little air through but keeps the queen from getting out.

Step 6: darkness

Wrap the tube in foil or black fabric. Ants found in total darkness (remember: in the wild they dig deep). Place the tube horizontally in a quiet spot at 20-25°C.

How long the tube lasts

A well-prepared tube stays functional for 3 to 6 months. Beyond that, the water runs out (check the level visually) or mould may appear. Prepare a fresh tube for rotation.
